Hi, guys. Since my background is physics, I have some simple questions about the homework:
1. It is said in the abstract of Monitoring of transcriptional regulation in Pichia pastoris under protein production conditions that "Fab secretion rates 1.9 fold." What does the unit "fold" mean?

2. In the abstract of Novel insights into the unfolded protein response using Pichia pastoris specific DNA microarrays, it is said that " a strain overexpressing the UPR transcription factor HACI". But how to overexpress a gene?

Sure, about your questions.

1. "1.9 fold" means "1.9 times the reference level," where the reference level is the amount produced by the wild type strain.

2. To overexpress Hac1 means that the strain expresses Hac1 more than the wild type.  The usual way this is done is to put the gene on a plasmid and then insert (transform) the plasmid into a cell.

As far as I can tell, there aren't really good metabolic models for pichia out there.  it would be very interesting if we could find a way to adapt a relevant cerevisiae one.
