Abstract: Single-molecule reconstruction by cryo-electron microscopy (cryo-EM) is an emerging approach to analyze structure of protein complex in its native state. In this presentation, we will discuss the current state of the art of the approach, its application to understand the molecular architecture of HIV-1 membrane envelope glycoprotein trimeric precursor (gp160), as well as the prospect of further development of the approach and its potential impact on molecular medicine in the near future. We will explain why the application of single-molecule cryo-EM to the case of analysis of a metastable architecture of HIV-1 gp160 trimer is unprecedented and likely leads to a revolution in modern structural biology and molecular medicine. Comments on the current debate and controversy in the area of cryo-EM methodology development and application will be given.
