摘要:

Intrinsically disordered proteins (IDPs) account for a significant fraction of any proteome and are central to numerous cellular functions. Yet how sequences of IDPs code for their conformational ensembles, conformational dynamics, and ultimately, functions is poorly understood. I will present advances in three areas using molecular dynamics simulations complemented by NMR spectroscopy. First, we have identified local interactions and residual secondary structure as major determinants of backbone dynamics and, based on this understanding, developed a predictor, SeqDYN, for backbone amide transverse relaxation rates. Second, we have elucidated a myriad of activities mediated by membrane association, including membrane anchoring of the Z-ring for bacterial cell division and facilitation of microtubule binding, as well as membrane-assisted aggregation. Lastly, we have demonstrated that residue-specific drug binding propensities of IDPs can be predicted from sequences.

Professor Huan-Xiang Zhou is the LAS Endowed Chair in Natural Sciences at UIC since 2017. He is a Fellow of the Biophysical Society (2023), American Physical Society (2010), and AAAS (2008). His research group focuses on theoretical, computational, and experimental studies in molecular and cellular biophysics.
