Single-molecule reconstruction by cryo-electron microscopy (cryo-EM) is anemerging approach to analyze structure of protein complex in its native state.In this presentation, we will discuss the current state of the art of theapproach, its application to understand the molecular architecture of HIV-1membrane envelope glycoprotein trimeric precursor (gp160), as well as theprospect of further development of the approach and its potential impact onmolecular medicine in the near future. We will explain why the application ofsingle-molecule cryo-EM to the case of analysis of a metastable architecture ofHIV-1 gp160 trimer is unprecedented and likely leads to a revolution in modernstructural biology and molecular medicine. Comments on the current debate andcontroversy in the area of cryo-EM methodology development and application willbe given.
