Single-molecule studies have revolutionized the field of molecular biology and cell biology due to their high spatial and temporal resolution. In the first part of the seminar, I will present my single-molecule fluorescence work on several prominent biological systems such as microRNA and CRISPR. In the second part, I will present very recent effort in my group on developing two next-generation single-molecule tools—making a first-ever massively parallel single-molecule fluorescence method that will allow for genome-wide single-molecule kinetic studies; and making single-molecule protein sequencers that have a potential for protein discovery and early disease detection. When these methods are further developed to study intrinsically complex biological phenomena, they will make breakthroughs in biology, biotechnology, and medical sciences.

报告人简介:

Chirlmin Joo obtained his PhD in physics at the University of Illinois at Urbana-Champaign. During his PhD study, he advanced single-molecule techniques by innovating new tools including single-molecule multi-color FRET. As a post-doctoral fellow at Seoul National University, he discovered novel proteins in microRNA biogenesis. As a research professor in the same institute, he succeeded reconstitution of functional protein complexes at the single-molecule level.

In 2011, he started his faculty position in Department of Bionanoscience at Delft University of Technology in the Netherlands. In 2017, Chirlmin Joo took leadership of developing new techniques of 'single-molecule protein sequencing' in a Dutch national science program (NWO-FOM Vrije Programme). The new technology will be the first to create the opportunity for single-cell proteomics and real-time screening for on-site medical diagnostics. In 2018, Chirlmin Joo was appointed Director of Kavli Institute of Nanoscience Delft.
